Fusarium wilt caused by Fusarium oxysporum f.sp. melonis (FOM) is a devastating disease of melon worldwide. Pathogenicity tests performed with F. oxysporum isolates obtained from Italian melon‐growing areas allowed to identify thirty‐four FOM isolates and the presence of all four races. The aims of this work were to examine genetic relatedness among FOM isolates by race determination and to perform phylogenetic analyses of identified FOM races including also other formae speciales of F. oxysporum of cucurbits. Results showed that FOM race 1,2 was the most numerous with a total of eighteen isolates, while six and nine isolates were identified as race 0 and 1, respectively, and just one isolate was assigned to race 2. Phylogenetic analysis was performed by random amplified polymorphic DNA (RAPD) profiling and by translation elongation factor‐1α (TEF‐1α) sequencing. The analysis of RAPD profiles separated FOM races into two distinct clades. Clade 1, which included races 0, 1 and 1,2, was further divided into ‘subclade a’ which grouped almost all race 1,2 isolates, and into ‘subclade b’ which included race 0 and 1 isolates. Clade 2 comprised only race 2 isolates. The phylogenetic analysis based on TEF‐1α separated FOM from the other formae speciales of F. oxysporum. Also with TEF‐1α analysis, FOM races 0, 1 and 1,2 isolates grouped in one single clade clearly separated from FOM race 2 isolates which grouped closer to F. oxysporum f.sp. cucumerinum. RAPD technique was more effective than TEF‐1α in differentiating FOM race 1,2 isolates from those belonging to the closely related races 0 and 1. Both phylogenetic analyses supported the close relationship between the three different FOM races which might imply the derivation from one another and the different origin of FOM race 2.  相似文献

The host suitability of five zucchini and three cucumber genotypes to Meloidogyne incognita (MiPM26) and M. javanica (Mj05) was determined in pot experiments in a greenhouse. The number of egg masses (EM) did not differ among the genotypes of zucchini or cucumber, but the eggs/plant and reproduction factor (Rf) did slightly. M. incognita MiPM26 showed lower EM, eggs/plant, and Rf than M. javanica Mj05. Examination of the zucchini galls for nematode postinfection development revealed unsuitable conditions for M. incognita MiPM26 as only 22% of the females produced EM compared to 95% of the M. javanica females. As far as cucumber was concerned, 86% of the M. incognita and 99% of the M. javanica females produced EM, respectively. In a second type of experiments, several populations of M. arenaria, M. incognita, and M. javanica were tested on zucchini cv. Amalthee and cucumber cv. Dasher II to assess the parasitic variation among species and populations of Meloidogyne. A greater parasitic variation was observed in zucchini than cucumber. Zucchini responded as a poor host for M. incognita MiPM26, MiAL09, and MiAL48, but as a good host for MiAL10 and MiAL15. Intraspecific variation was not observed among the M. javanica or M. arenaria populations. Cucumber was a good host for all the tested populations. Overall, both cucurbits were suitable hosts for Meloidogyne but zucchini was a poorer host than the cucumber.  相似文献

河套蜜瓜果实发育过程中糖积累与蔗糖代谢相关酶的关系   总被引:3,自引:0,他引:3  
以河套蜜瓜为试材,采用外部形态观测与内部生理指标测定相结合的方法,对其果实发育过程中果实生长模式以及果实中蔗糖、果糖、葡萄糖和淀粉含量以及蔗糖代谢相关酶活性进行测定,以揭示河套蜜瓜果实生长发育过程中糖的代谢积累与相关酶的关系.结果显示:(1)河套蜜瓜果实生长速率呈单"S"曲线,果实发育早期以积累葡萄糖为主,进入成熟期后蔗糖积累量迅速增加,最终由蔗糖和己糖共同构成果实品质.(2)在河套蜜瓜果实成熟期前,蔗糖磷酸合成酶(SPS)活性维持较低水平,进入成熟期后,SPS活性迅速升高;蔗糖合成酶(SS)活性在成熟期前为分解活性大于合成活性,成熟期后表现为合成活性大于分解活性;在整个果实发育期,酸性转化酶(AI)活性较低,中性转化酶(NI)活性始终高于AI.(3)在果实整个发育期,蔗糖含量与蔗糖代谢酶的净活力呈极显著正相关,蔗糖代谢相关酶共同作用决定果实中蔗糖含量.研究表明,在河套蜜瓜果实发育前期,以蔗糖分解代谢为主,且蔗糖合成酶和中性转化酶是催化蔗糖分解的关键酶;果实成熟期间,蔗糖代谢转为合成方向为主,蔗糖合成酶和蔗糖磷酸合成酶在蔗糖积累中起主导作用.  相似文献

以网纹甜瓜品种‘中蜜1号’为试材,在其不同培养阶段添加不同浓度AgNO3,检测不定芽的诱导率和增殖率、玻璃苗发生率和生根情况的结果表明,添加AgNO3后,子叶的愈伤组织诱导率下降,添加70μmol·L^-1AgNO3可促进不定芽的诱导,50~60μmol·L^-1AgNO3明显促进不定芽增殖,并完全抑制玻璃苗的产生,添加50μmol·L^-1AgNO3的生根最好。  相似文献

This study examined the host-selection ability of the broad mite Polyphagotarsonemus latus (Banks) (Acari: Tarsonemidae). To make long-distance-shifts from one host plant patch to another, broad mites largely depend on phoretic association with whiteflies. However, the host plants of whiteflies and broad mites are not necessarily the same. We determined the host-preference and acceptance of free-moving and phoretic broad mites using two behavioral bioassays. We used a choice test to monitor host selection by free-moving mites. In the case of phoretic mites, we compared their rate of detachment from the phoretic vector Bemisia tabaci placed on leaves taken from various host plants. The suitability of the plant was further determined by monitoring mite’s fecundity and its offspring development. We compared the mites’ responses to young and old cucumber (Cucumis sativus cv. ‘Kfir’) leaves (3rd and 8–9th leaf from the apex, respectively), and two tomato (Solanum lycopersicum cvs. ‘M82’ and ‘Moneymaker). Free-moving mites of all stages and both sexes preferred young cucumber leaves to old cucumber leaves and preferred young cucumber rather than young tomato leaves, demonstrating for the first time that broad mites are able to choose their host actively. As for phoretic mated females, although eventually most of the mites abandoned the phoretic vector, the rate of detachment from the whitefly vector was host dependent and correlated with the mites’ fitness on the particular host. In general, host preference of phoretic female mites resembled that of the free-moving female. Cues used by mites for host selection remain to be explored.  相似文献

采用营养液水培,研究了外源亚精胺(Spd)对NaCl胁迫下抗盐能力不同的两个黄瓜品种幼苗生长、叶绿体中活性氧清除系统、转谷酰胺酶(TGase)活性、结合态多胺含量及植株光合速率的影响.结果表明,外源Spd能提高NaCl胁迫下叶绿体中TGase活性、叶绿体结合态腐胺(Put)、Spd、精胺(Spm)及总多胺含量;提高超氧化物歧化酶(SOD)、抗坏血酸过氧化物酶(APX)和谷胱甘肽还原酶(GR)活性,提高抗坏血酸(AsA)、类胡萝卜素(Car)、还原型谷胱甘肽(GSH)含量及还原型谷胱甘肽/氧化型谷胱甘肽(GSH/ GSSG)比值,降低脱氢抗坏血酸/抗坏血酸(DAsA/AsA)比值;同时显著降低叶绿体过氧化氢(H2O2)和丙二醛(MDA)含量,提高植株净光合速率,缓解NaCl胁迫对幼苗生长的抑制.表明Spd对黄瓜盐害的缓解作用之一可能是通过提高叶绿体结合态多胺含量和叶绿体活性氧清除能力,从而缓解盐胁迫对叶绿体膜的伤害.  相似文献

The development of plants depends on the photoperiod length, light intensity, temperature, and length of light day integral. The reaction of a plant to the day length or daily light integral can depend on both the range of studied light intensities and photoperiod. Based on the data concerning the effects of light and thermal integrals on the developmental rate of plants of different photoperiodic groups, a photothermal model of plant development was proposed. The model was used to calculate the lengths of optimal photoperiods and ranges of daily temperature gradients ensuring the highest developmental rate of some plants, such as soybean, wheat, cucumber, and barley.  相似文献

采用丝氨酸内肽酶抑制剂和植物生长调节剂处理离体黄瓜叶片,研究了黄瓜叶片暗诱导衰老过程中丝氨酸内肽酶的作用。结果表明,6-BA50μmol/L与丝氨酸内肽酶抑制剂AEBSF能抑制叶片内肽酶活性的升高,延缓蛋白质降解,而ABA50μmol/L则促进了内肽酶活性的升高:其作用效果与AEBSF相反。活性电泳结果显示,黄瓜叶片中检测到6条内肽酶同工酶,其中4条(CEP2、3、4、6)为丝氨酸类型内肽酶,而ABA使丝氨酸内肽酶CEP2、3、4、6的活性明显增强,提示了丝氨酸类型内肽酶在黄瓜叶片衰老过程中具有重要作用。  相似文献

In order to clarify the response of antioxidant systems in various cellular organelles to photo-oxidative stress, the activities of superoxide dismutase (SOD) and enzymes of the ascorbate–glutathione (AsA-GSH) cycle were investigated in chloroplasts, mitochondria and cytosol of cucumber leaves subjected to methyl viologen (MV) treatment. Photo-oxidation by MV resulted in significant reductions in net photosynthetic rate (Pn) and increases in the ratio of the quantum efficiency of photosystem II (PSII), ΦPSII to that of the quantum efficiency of CO2 fixation (ΦCO2), followed by increased activities of SOD, and a general increase of AsA-GSH cycle enzymes in chloroplasts, mitochondria and cytosol. These increases were however, most significant in chloroplasts. There were also significant increases in dehydroascorbate (DHA), reduced glutathione (GSH), and oxidized glutathione (GSSG) except that the content of ascorbate (AsA) in chloroplasts and cytosol was slightly decreased and little effected, respectively. However, GSSG in mitochondria and GSH in cytosol were little influenced by the MV treatment. The activity of ascorbate oxidase (AO) in these organelles was independent of the MV treatment while the activity of l-galactono-1,4- lactone dehydrogenase (GLDH) in mitochondria was slightly inhibited by MV treatment. These results indicate that disturbance of electron transport in chloroplasts by MV influenced the metabolism of whole cell by a crosstalk signaling system and that the AsA-GSH cycle played a primary role in sustaining the levels of AsA.  相似文献

以薄皮甜瓜品种‘玉美人’作接穗,以白籽南瓜‘圣砧一号’为砧木进行嫁接,以自根苗为对照的结果表明,嫁接植株的南瓜根系主动吸收能力增强,其伤流量比自根植株的大,植株吸收氮钾的能力高于自根植株,而吸收磷的能力则有所降低。伤流液中玉米素(ZT)、赤霉素(GAs)和脱落酸(ABA)的浓度均低于自根植株,但ZT和GA的含量高于自根苗,而ABA的含量低于自根苗。嫁接植株的增产效果显著,其平均单瓜重和667m^2产量均高于自根植株。  相似文献
